Semaglutide / Tirzepatide

Struggling to slim down with diet and exercise alone? Medications like semaglutide and tirzepatide may help boost your weight loss efforts. These innovative prescription drugs work by mimicking the appetite-regulating hormone GLP1 to promote feelings of fullness and reduce hunger.

In clinical trials, weekly injections of semaglutide and tirzepatide have been shown to significantly decrease body weight compared to placebos and other anti-obesity medications. Participants also achieved improvements in related health parameters like waist circumference, BMI, blood pressure, blood lipids, and blood sugar levels.

Both semaglutide and tirzepatide seem to be well-tolerated overall, with the most common side effects being temporary GI issues like nausea, vomiting, and diarrhea. These usually resolve within a few weeks of starting treatment.

Tirzepatide has additional benefits over semaglutide for weight management and metabolic health. In head-to-head studies, tirzepatide outperformed semaglutide in average weight loss over the same period. Study participants on tirzepatide lost on average, between 12.4% (5mg dose) and 22.5% (15mg dose) of their body weight over 72 weeks. With semaglutide 2.4mg once weekly, participants achieved an average of 15.2% reduction in body weight.

Tirzepatide also showed greater improvements in cardiometabolic markers like waist circumference, triglycerides, HDL, and LDL cholesterol compared to semaglutide.

Researchers think tirzepatide’s dual mechanism of action on both GIP and GLP1 hormone pathways drives enhanced efficacy.

Both GLP1 drugs yield impressive results, but tirzepatide promotes greater weight loss and metabolic benefits overall. Still, semaglutide remains an excellent option for many individuals needing obesity pharmacotherapy.

Weight Loss Through Multiple Mechanisms

While effective for many, traditional lifestyle interventions often fail long-term due to biology working against us. Our bodies react to weight loss by increasing appetite and slowing metabolism to encourage regaining pounds for survival. GLP1 drugs help overcome this by influencing neurological signals, gastrointestinal processes, and more to facilitate fat burning.

First, GLP1s cross the blood-brain barrier to bind receptors that suppress appetite, reduce food motivation/cravings and improve satiety. This makes following a calorie-controlled diet much easier. They also slow stomach emptying and bowel motility, which stabilizes blood sugar and insulin while prolonging feelings of fullness after meals.

Additional proposed mechanisms driving weight loss include:

  • Increased thermogenesis and lipid metabolism – i.e. burning more calories
  • Improved glycemic control and insulin sensitivity
  • Protective effects on beta cell function and mass
  • Decreased fat absorption due to delayed gastric emptying
  • Direct and indirect effects on the gut microbiome

Through these synergistic actions on multiple bodily systems, GLP1s promote substantial slimming that diet and exercise alone rarely match. Their influence on numerous obesity-related comorbidities also leads to impressive cardiometabolic improvements.
